      <title>Cost and Processing Time Reductions for Insulin Purification Achieved with YMC-Triart Stationary Phase</title>

Due to the typical scale of insulin production and market pressures to increase availability and reduce the cost of insulin, there is justification to invest resources into finding improved conditions and materials for insulin purification.<br />

YMC participated in a detailed cost calculation analysis for insulation purification under the supervision and with the cooperation of a leading insulin manufacturer.<br />
<br />
The results of the study have been released by YMC in a case study entitled <a class="extlink"  target="_blank"  rel="nofollow noopener" title="How to reduce the costs for preparative LC processes" href="https://www.ymcamerica.com/wp-content/uploads/2021/04/Insulin-Purification-Case-Study.pdf">How to reduce the costs for preparative LC processes</a>.<br />

The case study points out that there are three cost drivers associated with preparative LC processes: labor/overhead comprises roughly 56 percent of the cost; mobile phase consumption accounts for 31 percent; and the stationary phase accounts for the remaining 13 percent — the smallest portion of the cost.<br />
<br />
Ironically, it is the stationary phase that can exert the most significant impact on the processing time (overhead) and mobile phase consumption — the lowest contributor to cost has the greatest impact on overall cost-effectiveness.<br />
<br />
In the case study, several stationary phases were tested (all of them C8 reversed-phase). Based on the tests, the study performed a careful cost estimation for the isolation of 100kg of purified insulin.<br />
<br />
Key factors in the cost projections included insulin recovery, the amount of crude insulin required to produce 100g of 99.5% pure product, the required number of purification cycles, fraction volume per run, the number of days required to reach the goal of 100g, and the total volume of solvent required.<br />
<br />
In the study, YMC-Triart Prep Bio200 C8 outperformed all other phases in the test, not just in overall projection but in each individual metric.<br />
<br />
YMC-Triart Prep Bio200 C8 accounted for the achievement of the goal of 100g of 99.5% pure insulin in the shortest period of time, with the lowest number of injections, the lowest amount of solvent required, and the smallest amount of crude insulin required. Overall, the projected savings in purification costs are up to 40 percent.<br />

Purification is a critical step in devising a cost-effective manufacturing process for peptide therapeutics — the goal being the optimized combination of loadability, recovery, and purity.<br />

The method for the purification of liraglutide (antidiabetic peptide marketed by Novo Nordisk as Victoza®) achieved high resolution using YMC-Triart Prep C18-S under alkaline conditions. The purity obtained for the target compound was 99.5 percent.<br />

Separation of the crude sample appeared to be successful at low to neutral pH, but a co-eluting peak emerged at pH 6.5, and the impurity was resolved at pH 8.5. <br />

Developing a method for peptide purification at pH 8.5 can be difficult due to the instability of typical reversed-phase media at a higher pH. YMC-Triart is a hybrid reversed-phase material that tolerates pH 8.5 (and higher) for the extended/continuous periods required in a manufacturing purification process. <br />

The Technical Note lists three critical contributing factors to the development of an efficient peptide purification process — Selectivity, Loadability/Recovery, and Media Lifetime (media stability) — and goes on to provide recommendations regarding each individual aspect.<br />

Selectivity determines the resolution of the separation. The highest purification productivity can be obtained only from a separation with optimal resolution. Therefore, says the Technical Note, it is important to screen different stationary phase chemistries under differing separation conditions.<br />

Loadability and Recovery are obviously at the heart of any purification method. Once again, screening of different stationary phases and variations in conditions will lead the analyst to the most suitable combination of media and conditions. For peptides, choosing a stationary phase with larger diameter pores (i.e., larger than 180A) will generally lead to optimal interaction between the peptide and the surface of the stationary phase. Too large a pore diameter, warns YMC, can lead to poor reproducibility.<br />

The goal of combining Selectivity and Loadability/Recovery is a result where the largest possible fractions can be concentrated and collected in the smallest possible volume, with the lowest contamination by adjacent fractions containing undesired material.<br />
<br />
The third contributor to successful peptide purification methods is Media Lifetime, or more accurately, useful Media Lifetime. The lifetime of a stationary phase has an obvious impact on the economics of the preparative purification process. The subtlety in this regard is that the stationary phase needs to remain stable and reproducible during its lifetime. Long life is not useful if the behavior of the material changes as a result of repeated use. <br />

Media Lifetime can be evaluated in terms of mechanical durability (stable when exposed to pressure shock and column re-packing cycles) and chemical stability (ability to withstand pH extremes during use and during alkaline cleaning in place (CIP) conditions).<br />

The lab is now readying to accept a production scale Contichrom TWIN 300 HPLC enabled with the patented multi-column countercurrent solvent gradient purification (MCSGP) technology. This twin-column HPLC system is the largest of its kind in the world and will be available for customer use as well as in-house development work. It joins the multiple Contichrom CUBE bench top units, which have the MCSGP functionality, already in use at the Kyoto Works facility. Uniquely, the unit is engineered to operate both as an HPLC designed to 100 bar as well as perform LPLC tasks such as <a class="extlink"  target="_blank"  rel="nofollow noopener" title="ion-exchange" href="https://www.ymcamerica.com/columns/ion-exchange/">ion-exchange</a> chromatography. A smaller-scale pilot GMP unit is available in the USA.<br />

Using the proprietary semi-continuous chromatographic process of MCSGP, users report 30-60% gains in yield while reducing solvent consumption up to 70% during purification of peptides and oligos. The MCSGP process works by automatically alternating the feeding of one of two identical columns while eluting the other column. The automatic internal recycling maintains the high product purity while purifying a much greater percentage of valuable product from a feed solution.<br />

In its inaugural year, the YMC Kyoto Works lab is operating in Phase I of a multiple phase roll out of capabilities. In Phase I, the facility has lab to mid capacity DAC (dynamic axial compression) column manufacturing (300~600?) and houses three (3) Contichrom CUBE instruments, an EcoPrime six (6) column simulated moving bed (SMB) HPLC, an SMB lab instrument, a Contichrom TWIN CaptureSMB 100 GMP scale LPLC, and a 1000?DAC system.<br />

The TWIN MCSGP unit adds its 0.3 to 3.3 LPM capacity to the already impressive array of technologies available in the YMC Kyoto Works. Phase II of Kyoto Works lab will see up to 200 employees with manufacturing capacity, and an additional building (explosion-proof/GMP) is planned for completion in 2021 on the 52,000 m<sup>2</sup> of YMC owned land.<br />

It is the seventh phase in the CHIRAL ART family of polysaccharide chiral phases for HPLC.<br />

Chiral phases are used for the separation of stereoisomers and enantiomers – analytes that are otherwise difficult to separate using widely used modes such as <a class="extlink"  target="_blank"  rel="nofollow noopener" title="reversed-phase" href="https://www.ymcamerica.com/columns/reversed-phase/">reversed-phase</a>, size exclusion, or ion exchange. But not all chiral phases perform well with all molecules of this type. Hence, access to a variety of chiral selectors is useful when working on a new separation challenge or improving an old one.<br />

The CHIRAL ART family of chiral phases includes five different chiral selectors. All five selectors – including Cellulose-SZ – are available as immobilized type; two selectors are also available in coated type.<br />

The immobilized type of chiral selector does not change the chemistry of the selector, but it does enable the use of a wider range of solvents, making them stable in both normal phase mode and reversed phase mode.<br />

The new CHIRAL ART Cellulose-SZ selector is cellulose tris(3-chloro-4-methylphenylcarbamate). Its stereoselectivity is complementary to the other cellulose tris and amylose tris selectors used in the other CHIRAL ART phases. <br />

CHIRAL ART Cellulose-SZ performs well in a wide range of pH conditions from pH 2 to pH 10. YMC-Triart Prep Bio200 C8 is an extension of the YMC-Triart product line that shares the fundamental advantages of the YMC-Triart hybrid base particle. YMC included certain product features designed specifically to improve results with insulin/peptide purification.<br />

The novel inorganic/organic hybrid silica is common to all the YMC-Triart phases. <br />

The flyer reviews the importance of the physical strength of YMC-Triart media. The results of a YMC study reveal little change in column pressure even after repeated packing-unpacking-repacking cycles. This is evidence of the mechanical durability of the material, as few fines are created during packing and unpacking.<br />

Dynamic compression applies pressure to the packed bed to keep it uniformly organized, but the pressure can damage – literally fracture – conventional silica-based phases. This stability facilitates the use of alkaline (or acidic) cleaning in place (CIP) protocols at extreme pH that would be too harsh for conventional media. The second characteristic is that YMC-Triart Prep Bio200 C8 has a particle pore size of 200A. Typical pore size for <a class="extlink"  target="_blank"  rel="nofollow noopener" title="reversed-phase" href="https://www.ymcamerica.com/columns/reversed-phase/">reversed-phase</a> media is 120A – this can be too small for good results with peptide molecules of 5-50 amino acids. On the other hand, a pore size of 300A, generally considered useful for proteins and intact antibodies, is larger than optimal for peptide purification.<br />

The analytes of interest are components of a commercial anti-itch medication. Separation under acidic or neutral conditions proved difficult, due to either co-elution or poor resolution.<br />

YMC&apos;s application shows optimal peak separation and peak shape using YMC-Triart <a class="extlink"  target="_blank"  rel="nofollow noopener" title="C18 Hybrid Column" href="https://www.ymcamerica.com/microcapillary-hplc/ymc-triart-c18/">C18 Hybrid Column</a> under alkaline (pH 9.6) gradient mobile phase conditions.  Further study resulted in an isocratic solution, also at pH 9.6. The new stationary phase – under the name YMC-Triart Prep Bio200 C8 – is an extension of the YMC-Triart product line that inherits the fundamental advantages of the YMC-Triart hybrid base particle. All YMC-Triart phases begin with the same type of novel inorganic/organic hybrid silica. This hybrid particle technology offers two important advantages to users involved in small or large production scale peptide purification.<br />

First, every YMC-Triart stationary phase, including YMC-Triart Prep Bio200 C8, is mechanically stable and physically strong. This means that the media can be expected to endure repeated column packing, unpacking, and re-packing cycles with minimal particle fracturing. Particle fractures (fines) are common with conventional silica-based media, contributing to increased back-pressure, and sometimes creating voids in the packed bed of media inside the column. YMC-Triart media overcome the weakness that is the principal cause of particle fracturing. <br />
<br />
The physical strength of YMC-Triart media also leads to performance improvements in the use of dynamic <a class="extlink"  target="_blank"  rel="nofollow noopener" title="compression columns" href="https://www.ymcamerica.com/ymc-dynamic-axial-compression-dac-columns-for-preparative-scale-liquid-chromatography/">compression columns</a> (most typically, dynamic axial compression (DAC) columns). Dynamic compression applies pressure to the packed bed to keep it uniformly organized. Bed uniformity is critical to the sharpness of eluted fractions and reducing fraction volume.<br />
<br />
The second important advantage of YMC-Triart Prep Bio200 C8 is its chemical stability. The media is stable at any pH from 1 to 10 for regular use and up to pH 12 for clean-in-place.  This stability enables the use of alkaline (or acidic) cleaning in place (CIP) protocols at extreme pH that would be too harsh for conventional media. The use of CIP protocols is important in peptide purification to reverse the deterioration of media performance caused by the adsorption of proteins.  <br />

YMC also incorporated two important adjustments to the properties of YMC-Triart Prep Bio200 C8. <br />
<br />
By using shorter chain C8 (USP designation L7) stationary phase chemistry, YMC-Triart Prep Bio200 C8 is measurably less retentive than C18 (ODS) materials often used for peptide purification. This has the effect of shortening fraction elution times in the peptide purification process.<br />
<br />
A second adjustment is that YMC-Triart Prep Bio200 C8 has a particle pore size of 200A. Typical pore size for <a class="extlink"  target="_blank"  rel="nofollow noopener" title="reversed-phase" href="https://www.ymcamerica.com/columns/reversed-phase/">reversed-phase</a> media is 120A. By fabricating YMC-Triart Prep Bio200 C8 with a 66% larger pore size, the media enables more consistent media surface interaction by peptide molecules of 5-50 amino acids. Consistent and reproducible media interaction is a key factor contributing to low volume elution of purified fractions. A pore size of 300A, generally considered useful for proteins and intact antibodies, is larger than optimal for peptide purification.<br />
